Immunogen

Recombinant human monocyte chemotactic protein 2 (MCP2), expressed in E. coli.

Localization

Specificity

By ELISA, this antibody shows cross-reactivity with recombinant human MCP1 and recombinat mouse MIP1 alpha. In addition, it does not cross-react with other chemokines tested: rmC10, rhENA78, rhGRO alpha, rhGRO beta, rhGRO gamma, rmJE, rhI309, and rhIL8. This antibody neutralizes the biological activity of recombinant human MCP-2. Shows

Applications for CCL8/MCP-2 Antibody (35509.11)

Application
Recommended Usage

ELISA

1 - 2 ug/ml

Immunohistochemistry-Paraffin

1:500-1:1000

Western Blot

1 - 2 ug/ml

Neutralization

1 - 4 ug/ml
Application Notes
ELISA: Use at a concentration of 0.5 - 1 ug/ml to detect 12.5ng/ml of recombinant human MCP2.Neut: Use at a concentration of 0.5 - 10 ug/ml. WB: Use at a concentration of 1 - 2 ug/ml to detect 50ng/lane of recombinant human MCP2 under reducing and non-reducing conditions. Predicted molecular weight: 9 kDa. Human MCP2 is tested for its ability to neutralize the chemotactic activity of rhMCP2 for human monocytes. The ND50 of the antibody is defined as the concentration of antibody resulting in a one-half maximal inhibition of bioactivity of rhMCP2 that is present at a concentration just high enough to elicit a maximum response. In this bioassay 0.4ug/ml rhMCP2 was mixed with various dilutions of the antibody for 30 minutes at 22.

Use in Immunocytochemistry/immunofluorescence reported in scientific literature (PMID : 28894190).
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: CCL8/MCP-2

MCP 2 (Monocyte Chemoattractant Protein 2) is a chemotactic factor that attracts monocytes, lymphocytes, basophils and eosinophils. MCP 2 is important to inflammatory host responses, and is found in the highest concentration in the small intestine and peripheral blood cells. This protein is structurally related to the CXC subfamily of cytokines. Members of this subfamily are characterized by two cysteines separated by a single amino acid. By recruiting leukocytes to sites of inflammation this cytokine may contribute to tumor-associated leukocyte infiltration and to the antiviral state against HIV infection.
