Why Culture Cardiomyocytes in a Defined Medium?
Maintaining the health of pluripotent stem cell-derived cardiomyocytes is imperative for generating trustworthy data during downstream experimentation. Culturing cardiomyocytes in serum-free, defined media reduces experimental variability in cell health by eliminating the uncontrolled variables that are often associated with serum-containing media, such as indeterminate levels of vitamins, hormones, and growth factors.
Cardiomyocytes cultured using the Cardiomyocyte Maintenance Media Supplement can also be a part of drug toxicity and small molecule screening workflows where a controlled cell culture environment is essential.
Cardiomyocyte Maintenance Media Use
Human pluripotent stem cells are differentiated into cardiomyocytes using the StemXVivo® Cardiomyocyte Differentiation Kit (R&D Systems, Catalog # SC032B) until widespread beating is observed in the culture. StemXVivo® Cardiomyocyte Maintenance Media can then be used to maintain the differentiated cells for extended culture. Media should be replaced daily.
Note: Repeated mechanical stress can negatively affect cell attachment and survival. For extended culture, daily media exchanges should be handled as gently as possible.
Precautions
This product contains human transferrin. This transferrin was purified from donor plasma and tested at the donor level using an FDA licensed method and found to be non-reactive for anti-HIV-1/2 and Hepatitis B surface antigen.
Embryonic stem (ES) cells are pluripotent stem cells derived from the inner cell mass of pre-implantation embryos. Induced pluripotent stem (iPS) cells can be generated by somatic cell reprogramming following the exogenous expression of specific transcription factors (Oct-3/4, KLF4, SOX2, and c-Myc). These cell types are capable of unlimited, undifferentiated proliferation in vitro and still maintain the capacity to differentiate into a wide variety of somatic cells. In this capacity, pluripotent stem cells have widespread clinical potential for the treatments of heart disease, diabetes, spinal cord injury, and a variety of neurodegenerative disorders.
R&D Systems offers a wide range of products to support pluripotent stem cell culture and differentiation. Mouse embryonic fibroblasts may be used to maintain and expand pluripotent stem cells in an undifferentiated state. We also offer defined culture media, which are specifically optimized for use with human or rodent pluripotent stem cells. Briefly, Cardiomyocyte Maintenance Media is prepared using the following procedure:
- Dilute 50-fold in RPMI 1640
- Add 0.5 mM L-glutamine and 1X Penicillin / Streptomycin (optional)
- Store the media in the dark at 2-8 °C
- Use within 2 weeks

Dilute 50-fold in RPMI 1640. Add 0.5 mM L-glutamine and 1X Penicillin / Streptomycin (optional). Store the media in the dark at 2 to 8 °C for up to two weeks.
