What is Conjugation?

Conjugation, also called labeling or tagging, refers to the process by which a molecule, such as a fluorophore, biotin, or enzyme, is attached to biomolecule, such as an antibody or protein. Conjugated proteins and antibodies are used in a variety of applications including flow cytometry, cytometry by time-of-flight (CyTOF), immunohistochemistry (IHC), immunocytochemistry (ICC), ELISA, and western blot (WB).

Labeling antibodies and proteins with biotin leverages one of the strongest non-covalent bonds in nature between biotin and streptavidin. Streptavidin has the capacity to bind four biotin molecules, which can dramatically enhance detection of low-abundance proteins. Amine- or Sugar-Labeled Biotinylated Proteins: Traditional biotinylation, using amine- or sugar-labeled biotinylation, typically incorporate multiple biotins per molecule, which can be advantageous for increasing signal strength. Similar to Avi-tagged biotinylated proteins, amine- and sugar-labeled biotinylated proteins are tested for lot-to-lot consistency and bioactivity following biotinylation to ensure the activity of the biotinylated protein is the same as the non-biotinylated protein. Proteins: HIS-tagged proteins are frequency used to aid in purification and protein detection in many different assays. Other tags, such as GST, HA, and Myc can be used for application-specific purposes.
