Background: 5'-Nucleotidase/CD73

CD73 is a membrane-bound enzyme having a key role in extracellular ATP metabolism producing adenosine by dephosphorylating AMP. It is highly expressed on endothelial cells but a moderate expression is also observed on a subpopulation of lymphocytes, especially regulatory T cells. Although CD73 on the endothelial cells and lymphocytes are structurally similar, they are differently regulated. Triggering of lymphocyte CD73 with antibodies results in cleavage of CD73 and activation of LFA-1, leading to increased adhesion of lymphocytes to endothelium (Algars A et al., 2011). CD73 plays an important role in carcinogenesis, as adenosine promotes tumor progression and CD73-expressing cancer cell lines are more aggressive (Supernat A et al., 2012). Extracellular adenosine is a potent immunosuppressor that accumulates during tumor growth. Anti-CD73 monoclonal antibody therapy significantly delays primary 4T1.2 and E0771 tumor growth in immune-competent mice and significantly inhibits the development of spontaneous 4T1.2 lung metastases. Antibody therapy is essentially dependent on the induction of adaptive anti-tumor immune responses (Stagg J et al., 2010).
