Background: ABCA7

ABCA7 (ATP-binding cassette sub-family A member 7) belongs to "A" subfamily of ABC transporters that predominantly transport lipids and other lipophilic molecules across membranes. ABCA7 shows higher expression in brain, lung, adrenal gland and spleen as well as hematopoietic tissues. It localizes to cell membrane ruffles/phagocytic cups of activated macrophages; to cytoplasm (Golgi and endosomes) of resting macrophages, and to the apical brush border of cells in the proximal tubules of kidney. It is also detected in both the plasma membrane and in intracellular compartments in myelo-lymphatic tissues (peripheral leukocytes, thymus, spleen, and bone marrow). ABCA7 facilitates the transport of phospholipids and, to a lesser extent, cholesterol, across cell membranes to extracellular apoAI and apoE discoidal lipoprotein complexes. Moreover, it implicates in macrophages mediated phagocytosis of apoptotic cells, and binds APOA1 for involving in apolipoprotein-mediated phospholipid efflux from cells. ABCA7 also mediate cholesterol efflux, and can regulate the cellular ceramide homeostasis during keratinocytes differentiation.
