Background: AIP/ARA9

AIP/ARA9 (AhR-interacting protein /AhR-associated protein 9) is a 37 kD immunophilin-like factor expressed ubiquitously with considerable variation in intracellular levels between different tissues. Its high levels of expression are observed in spleen, thymus, pituitary heart, placenta and skeletal muscle, whereas low expression levels are seen in liver, kidney and lung. AIP/ARA9 plays a positive role in AHR-mediated signaling, possibly by influencing its receptivity for ligand and/or its nuclear targeting. It was originally identified as a negative regulator of hepatitis B virus X-associated protein (hence named XAP2 also) and later was shown as Hsp90-associated protein which specifically interacts with AhR leading to regulation of AhR intracellular localization as well as protein stability via inhibition of AhR ubiquitination. AIP/ARA9 was also shown to interact with other signal transduction proteins like Ga13 and nuclear receptor transcription factors including GR, TR-beta1 and PPAR-alpha. Germline mutations that disrupt AIP/ARA9 have been reported in both familial as well as sporadic pituitary tumors and targeted disruption of this protein in mouse leads to cardiac malformation and embryonic lethality. Defects in AIP/ARA9 are the cause of growth hormone-secreting pituitary adenoma (GHSPA), ACTH-secreting pituitary adenoma (ASPA) and prolactin-secreting pituitary adenoma (PSPA).
