In Western blot analysis, a band is seen at ~20 kDa. There is also a band of greater intensity at ~40 kDa that may be an aggregated heteromeric complex. **CryAB aggregates with homologous proteins, including CryAA and the small heat shock protein HSP28, to form large heteromeric complexes.

Background: AlphaB Crystallin/CRYAB

Crystallin AB (CRYAB or alpha-basic-crystallin) belongs to HSP20 family and was originally discovered as chaperone abundantly expressed in eye lens where it plays a key role in maintaining transparency and refractive index of lens. Subsequently, CRYAB was found to acts as a molecular chaperone induced by a variety of stress stimuli, conferring cytoprotection by suppressing aggregation of denatured protein. In its anti-apoptotic role, CRYAB protects cells against stress, TNF-alpha, apoptotic effectors etc. and can inhibit caspase-3/PARP alongwith preventing Bax/Bcl-2 translocation. CRYAB is a heteropolymer of three CRYAA and one CRYAB subunits, and can aggregate with homologous proteins (e.g. HSPB1) to form large complexes and interact with HSPBAP1 as well as TTN/titin. Localized in cytoplasm, during heat shock stimulus, CRYAB translocates to sub-nuclear structures known as SC35/nuclear splicing speckles. CRYAB is expressed in several other tissues such as muscle, brain, spleen, lung, kidney, skin etc and has been shown to implicate in certain diseases including cardiac myopathies, neurodegenerative diseases and malignancies including brain, breast, prostate and renal cancers. Defectsive CRYAB causes myopathy myofibrillar type 2 (MFM2), cataract posterior polar type 2 (CTPP2) and myopathy myofibrillar fatal infantile hypertonic alpha-B crystallin-related (MFMFIH-CRYAB).
