Background: BAI1

BAI1 (brain-specific angiogenesis inhibitor 1) belongs to GPCR2 family or so-called adhesion-type family of 7-transmembrane receptors, and an extracellular fragment of this receptor was originally discovered to inhibit neovascularization in brain tumor model. BAI1 is a multi-pass membrane protein localized in cell membranes and is specifically expressed in brain with reduced or no expression in certain glioblastoma cell lines/cancer tissues and astrocytes. BAI1 is expressed on macrophages also, where it acts as a receptor for clearance of apoptotic cells via binding to surface phosphatidylserine of apoptotic cells through its type I TSRs followed by triggering their engulfment via an ELMO1/DOCK180/Rac1-mediated signaling pathway. BAI1 interacts with several proteins including MAGI1, MAGI3, BAIAP2, PHYHIP, ELMO1 and DOCK1. BAI1 plays a unique function in binding and internalization of Gram-negative bacteria through recognition of bacterial LPS via BAI1-TSRs. It has been proposed as a potent inhibitor of brain angiogenesis, a mediator of p53 signal in suppression of glioblastoma, and functions in cell adhesion as well as signal transduction in brain. BAI1 also regulates phagocytosis, synaptogenesis, inhibition of tumor growth and angiogenesis via processing of its extracellular domain into secreted vasculostatins, and it is emerging as a therapeutics target for diseases such as cancer, stroke, and schizophrenia.
