ELISA: Use at a concentration of 0.5 - 1.0 ug/ml. The detecton limit for recombinant human BMP-6 is approximately 6.2 ng/well.Inhib: Use at a concentration of 1 - 3 ug/ml.WB: Use at a concentration of 1 - 20 ug/ml. The detection limit for recombinant human BMP6 is approximately 20 ng/lane under non-reducing conditions.Anti-Human Bone Morphogenetic Protein 6 (BMP6) is measured by its ability to neutralize the bioactivity of BMP6 in osteoblast MC3T3-E1 cells. Recombinant human BMP6 is added to various concentrations of the antibody for 1 hour at 37. This antibody has neutralizing activity at 0.5-2 ug/ml in an assay using ATDC-5 cells, which indicates that it has specificity to an epitope of the target which neutralizes its functional activity.

Background: BMP-6

Bone Morphogenetic Proteins (BMP) are members of the TGF beta superfamily of cytokines that affect bone and cartilage formation. Similar to other TGF beta family proteins, BMPs are highly conserved across animal species. BMPs are involved in embryogenesis and morphogenesis of various tissues and organs. BMP6 is an autocrine stimulator of chondrocyte differentiation and has been implicated in the development of embryonic kidney and urinary systems. It is involved in liver growth and differentiation, keratinocyte differentiation, and regulation of neuronal tissue development. BMP6 expression is localized to smooth muscle cells of normal cerebral blood vessels, intimal plaques and atherosclerotically changed blood vessels, further suggesting a role as a regulator of developing tissues.
