Competitive ELISA, ELISA, Radioimmunoassay

Label

Unconjugated

Antibody Source

Monoclonal Mouse IgG1 kappa Clone # 7E10

Concentration

Concentrations vary lot to lot. See vial label for concentration. If unlisted please contact technical services.

Product Specifications

Immunogen

Derived from hybridization of X63-Ag8-653 myeloma cells with spleen cells of Balb/c mice immunized with recombinant C-peptide construction.

Reactivity Notes

No cross-reactivity with bovine, porcine and mouse/rat insulin.

Specificity

This is reacting only with human C-peptide. Binds to the human free C-peptide and C-peptide region in proinsulin molecules. Antigenic determinants identified by monoclnal antibodies are in the three-dimensional structure generated by N- and C- terminal regions of C-peptide separated by beta-turn at position 47-50 of proinsulin.

Monoclonal

Host

Mouse

Isotype

IgG1 kappa

Applications for C-Peptide Antibody (7E10)

Application
Recommended Usage

ELISA

1:100-1:2000

Radioimmunoassay

1:100-1:2000
Application Notes
Competitive ELISA or RIA detection of C-peptide in human serum after immunospecific removal of proinsulin (with solid-phased MAb 1D6 to human proinsulin (Cat.# NB110-2556)). Construction of a sensitive two-site sandwich EIA/ RIA immunosorbent assay for the measurement of total human proinsulin (including proinsulin convertion intermediates) in serum. Specific immunosorbtional removal of total proinsulin and C-peptide molecules from serum samples (with solid-phase 1H8, 2B7 and 4H8) before the measurement of human insulin.

Background: C-Peptide

C Peptide is part of the molecule of Proinsulin, that consists of three parts: C Peptide and two long strands of amino acids (called the alpha and beta chains) that later become linked together to form the insulin molecule. From every molecule of proinsulin, one molecule of insulin plus one molecule of C Peptide are produced. C peptide is released into the blood stream in equal amounts to insulin. A test of C peptide levels will show how much insulin the body is making. Insulin decreases blood glucose concentration. It increases cell permeability to monosaccharides, amino acids and fatty acids. It accelerates glycolysis, the pentose phosphate cycle, and glycogen synthesis in liver.
