Application Notes
Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 10 mM sodium citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.

Background: CCR6

CD196 (CCR6) is a chemokine receptor expressed on Th17 cells and regulatory Tcells that is induced by T-cell priming with certain cytokines. This protein plays a pivotal role in directing inflammatory and regulatory cells to inflamed tissues or the gut, where the ligand of CD196, CCL20 is expressed. Although most chemokine-chemokine receptor interactions are characterized by a high promiscuity, CD196 binds only CCL20 and -defensins. This chemokine receptor is widely expressed on human blood and tissue leukocytes, including subsets of dendritic cells, CD45RO+ effector/memory Tcells, CD25 high regulatory Tcells (Tregs), naive and memory B cells, NKT cells, and NK cells. On memory Tcells, CD196 is expressed on a population of autoreactive interleukin-10 (IL-10)-producing cells and subsets of skin- and mucosa-homing cells. More recently, CD196 was identified as a new risk factor for rheumatoid arthritis; moreover, a nucleotide polymorphism within the Cd196locus was shown to be associated with susceptibility to rheumatoid arthritis, Graves disease, and Crohn disease (Steinfelder S et al, .2011).This protein also acts as a marker for Th17 lineage polarization and HIV permissiveness in memory CD4+ Tcells. CD196+ Tcells have the potential to migrate into the GALT via the gut-homing integrin 47, a newly identified HIV-gp120 binding receptor. Nevertheless, given the ability of integrin 7 to regulate cell migration into the GALT and bind HIV-gp120, CD196+ Tcells coexpressing integrin 7 and CCR5 might have an extraordinary ability to disseminate HIV from the portal sites of entry (Monteiro P et al ,. 2011). Anti-CD196 antibody could almost completely suppress the cell migration induced by hBD-2 (Human beta-defensin-2), suggesting that hBD-2 mainly utilizes CD196 (CCR6) as a functional receptor (Niyonsaba F et al,.2004).
