Description

This antibody can be stored at 2C-8C for one month without detectable loss of activity. Antibody products are stable for twelve months from date of receipt when stored at -20C to -80C. Avoid repeated freeze-thaw cycles.

Immunocytochemistry/Immunofluorescence: CD117/c-kit Antibody (007) [NBP2-89411] - Staining of kit in HESS9 cells. Cells were fixed with 4% PFA, blocked with 10% serum, and incubated with rabbit anti-human kit monoclonal antibody (1:60) at 4c overnight. Then cells were stained with the Alexa Fluor(R) 488-conjugated Goat Anti-rabbit IgG secondary antibody (green).

Flow Cytometry: CD117/c-kit Antibody (007) [NBP2-89411] - Analysis of Human KIT(CD117) expression on TF-1 cells. Cells were stained with purified anti-Human KIT(CD117) (Filled hisgram), then a FITC-conjugated second step antibody. To demonstrate specificity of staining, the binding by was blocked by preincubation of the purified antibody with 20ug recombinant human KIT(CD117) for 1 hour (Black solid line hisgram). The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact cells.

Background: CD117/c-kit

CD117 is a 145 kD immunoglobulin superfamily member, also known as c-Kit and stem cell factor receptor (SCFR). It is a transmembrane tyrosine-kinase receptor that binds the c-Kit ligand (also known as steel factor, stem cell factor, and mast cell growth factor). CD117 is expressed on hematopoietic stem cells (including multipotent hematopoietic stem cells, progenitors committed to myeloid and/or erythroid lineages, and T and B cell precursors), mast cells, and acute myeloid leukemia (AML) cells. CD117 interaction with it's ligand is critical for the development of hematopoietic stem cells. The 2B8 antibody does not block c-Kit activity.
