Immunohistochemistry: CD30/TNFRSF8 Antibody (Ki-3) - Chimeric [NBP2-52682] - Immunohistochemical staining of human tonsil tissue using anti-CD30 antibody. Ki-3 Anti-CD30 staining of paraffin embedded human tonsil tissue using the rabbit-chimeric version of Ki-3 (NBP2-52682). Antigen retreival was acheived by microwaving in citrate buffer, followed by blocking with protein block serum-free buffer. Primary antibody incubation with NBP2-52682 was carried out at 4 ug/ml for 30 minutes. Samples were then incubated with an anti-rabbit IgG HRP secondary antibody for 20 mins followed by DAB (3,3'-diaminobenzidine), and counter-staining with haemotoxylin. Staining of the cellular membrane of dividing cells may be observed. Recommended concentration, 2-4 ug/ml.

Flow Cytometry: CD30/TNFRSF8 Antibody (Ki-3) - Chimeric [NBP2-52682] - Flow-cytometry using the anti-CD30 (Tumor necrosis factor receptor superfamily member 8) antibody Ki-3. Jurkat cells were stained with unimmunized rabbit IgG antibody (black line) or the rabbit-chimeric version of Ki-3 (NBP2-52682, blue line) at a concentration of 10 ug/ml for 30 mins at RT. After washing, bound antibody was detected using an anti-rabbit IgG JK (FITC-conjugate) antibody at 2 ug/ml and cells analyzed on a FACSCanto flow-cytometer.

Application Notes
This antibody is Cytof ready. This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG2b format, for improved compatibility with existing reagents, assays and techniques. The antibody binds specifically to CD30/TNFRSF8 a cell membrane protein which belongs to the tumour necrosis factor receptor family. Incubation of CD30/TNFRSF8 positive L540 cells with Ki-3 antibody weakly enhances release of soluble CD30/TNFRSF8 (in contrast to Ki-4, NBP2-52714). CD30/TNFRSF8 is expressed on activated lymphocytes, and binding by its ligands TRAF2 and TRAF5 results in the activation of NF-kappaB, thereby regulating gene expression. CD30/TNFRSF8 is also associated with Hodgkin Lymphoma, and its expression acts as a tumour marker. Targeting the CD30/TNFRSF8 antigen with specific monoclonal antibodies is a possible therapy for Hodgkin's lymphoma.

Background: CD30/TNFRSF8

CD30 is a type I transmembrane protein and a member of the tumor necrosis factor receptor superfamily of proteins. (1,2) Murine CD30 is expressed predominantly in the thymus, and it is inducible in mouse splenocytes stimulated with pokeweed mitogen or concanavalin A. (1) In anti-CD3epsilon-activated spleen cells, CD30 is expressed primarily on the surface of CD8+ T cells with peak expression on days 4 and 5. Stimulation of CD30+ CTL lines with plate-bound anti-CD30 directly signals IL-5 but not IFN; production. (1) While these studies demonstrate that CD30 directs cytokine secretion and suggest that CD30 may play a pivotal role in the pattern of cytokine production by T cells, the precise roles of CD30 and its ligand (CD153) in T-cell development have not been clearly defined.
