Western Blot: CD7 Antibody (3A1E) [NBP2-81050] - Western Blot using anti-CD7 antibody (3A1E) [NBP2-81050]. Jurkat cell lysate samples (35ug protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with the chimeric rabbit version of 3A1E [NBP2-81050] at 1 ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for unmodified CD7 is 25.4kDa, but this protein is glycosylated at several residues. NBP2-81050 successfully detected CD7 in Jurkat cell lysate.

Immunocytochemistry/Immunofluorescence: CD7 Antibody (3A1E) [NBP2-81050] - Immunofluorescence staining of fixed Jurkat cells with anti-CD7 antibody (3A1E) [NBP2-81050]. Immunofluorescence analysis of paraformaldehyde fixed Jurkat cells on Shi-fix coverslips, permeabilized with 0.15% Triton and stained with the chimeric rabbit IgG version of 3A1E [NBP2-81050] at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (1 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81050, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody followed by Alexa Fluor 488 secondary antibody.

Background: CD7

CD7, also known as gp40, is a 40 kD type I transmembrane glycoprotein member of the immunoglobulin superfamily. It is found on T cells, NK cells, thymocytes, hematopoietic progenitors, and monocytes (weakly). CD7 is also expressed on acute lymphocytic leukemia (ALL). CD7 crosslinking induces a calcium flux in T lymphocytes, presumably as a result of cytoplasmic domain association with PI3-kinase. CD7 co-stimulation can induce cytokine secretion and modulate cellular adhesion.
