Immunocytochemistry/Immunofluorescence: CD81 Antibody (1D6) [NBP2-81096] - Immunofluorescence analysis of paraformaldehyde fixed Jurkat cells on Shi-fix coverslips, permeabilized with 0.15% Triton and stained with the chimeric mouse IgG1 version of 1D6 at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81096, DAPI, merged channels and an isotype control. Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques. This antibody is a strong inducer of homotypic adhesion in hematolymphoid cells (Levy, 1998). This antibody also has an anti-proliferative effect, and is able to induce growth inhibition of B cell lines. Triggering of CD81 by this antibody induces a significant upregulation of the constituitive release of TNF-alpha by JY cells, an Eptsein-Barr virus-positive B lymphoblastoid cell line (Altomonte, 1995).

Background: CD81

CD81/TAPA-1 is an integral membrane protein expressed on a variety of cell types, and has a high degree of sequence homology between human and mouse. (1) CD81 is expressed on thymic stromal cells where it plays an important role in the transition of gamma delta + T cells to more mature T cells with alpha beta T cell receptors. (2) Immunohistochemical staining has revealed that its expression is localized to the subcapsular region of the thymus and, specifically, on cells that have distinct clustering patterns. It has been speculated that the ligand for CD81 is the pre-T cell receptor, which is composed of a TCR beta chain and glycoprotein pT alpha. (2) The monoclonal antibody 2F7 can block thymocyte interaction with CD81 in vitro. (2)
