Description

This antibody is an IgG fraction antibody purified from monospecific antiserum by a multi-step process which includes delipidation, salt fractionation and ion exchange chromatography followed by extensive dialysis against the buffer stated above. Assay by immunoelectrophoresis resulted in a single precipitin arc against anti-Rabbit Serum as well as purified and partially purified Elastase [Porcine Pancreas]

Store vial at 4C prior to restoration. For extended storage aliquot contents and freeze at -20C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4C as an undiluted liquid. Dilute only prior to immediate use.

This product has been tested by ELISA, western blot, and immunofluorescence. This product is assayed against 1.0 ug of Elastase [Porcine Pancreas] in a standard ELISA using Peroxidase conjugated Affinity Purified anti-Rabbit IgG [H&L] (Goat) and (ABTS (2,2'-azino-bis-[3-ethylbenthiazoline-6-sulfonic acid]) as a substrate for 30 minutes at room temperature. A working dilution of 1:1,000 to 1:5,400 is suggested for This product An investigator has reported that this antibody works very well on sections of mouse embryonic pancreas tissue, specifically staining developing exocrine cells.

Background: CELA1

Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, pancreatic elastase 1 is not expressed in the pancreas. To date, elastase 1 expression has only been detected in skin keratinocytes. Clinical literature that describes human elastase 1 activity in the pancreas is actually referring to elastase 2A.
