Western Blot: COASY Antibody (JE58-56) [NBP3-32203] - Western blot analysis of COASY on HepG2 cell lysates with Rabbit anti-COASY antibody (NBP3-32203) at 1/500 dilution.

Lysates/proteins at 10 ug/Lane.

Predicted band size: 60 kDa
Observed band size: 60 kDa

Exposure time: 2 minutes;

10% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32203) at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:200,000 dilution was used for 1 hour at room temperature.

Flow Cytometry: COASY Antibody (JE58-56) [NBP3-32203] - Flow cytometric analysis of COASY was done on HepG2 cells. The cells were fixed, permeabilized and stained with the primary antibody (NBP3-32203, 1ug/ml) (red) compared with Rabbit IgG, monoclonal - Isotype Control (green). After incubation of the primary antibody at +4℃ for 1 hour, the cells were stained with a Alexa Fluor®488 conjugate-Goat anti-Rabbit IgG Secondary antibody at 1/1,000 dilution for 30 minutes at +4℃ (dark incubation).Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

Background: COASY

Biosynthesis of coenzyme A (CoA) from pantothenic acid (vitamin B5) is an essential universal pathway in prokaryotes and eukaryotes. COASY is a bifunctional enzyme that catalyzes the 2 last steps in CoA synthesis. These activities are performed by 2 separate enzymes, phosphopantetheine adenylyltransferase (PPAT; EC 2.7.7.3) and dephospho-CoA kinase (DPCK; EC 2.7.1.24), in prokaryotes (Daugherty et al., 2002 [PubMed 11923312]).[supplied by OMIM]
