Product Summary for Crk [p Tyr251] Antibody

Immunogen

Phospho-Crk (Tyr-251) synthetic peptide (coupled to KLH) corresponding to amino acid residues surrounding Tyr-251 in human Crk II. This peptide sequence is well conserved in mouse and rat Crk II, as well as in CrkL (Tyr-251). The site is not found in Crk I. [UniProt# P46108]

Specificity

The antibody detects a 38 and 40 kDa* proteins corresponding to the molecular mass of Crk II and CrkL on SDS-PAGE immunoblots of human K562 cells stimulated with pervanadate and A431 cells stimulated with EGF. This reactivity is not observed after akaline phosphatase treatment.

Western Blot: Crk [p Tyr251] Antibody [NBP3-23255] - Western blot of human K562 cells stimulated with pervanadate (1 mM) for 30 min. (lanes 1-6). The blot was treated with akaline phosphatase to dephosphorylate Crk (lanes 2, 4 & 6), then the blot was probed with rabbit polyclonals CrkL (C-terminus) NBP3-23256 (lanes 1 & 2), Crk (Tyr-251) phospho-specific NBP3-23255 (lanes 3 & 4), and mouse monoclonal anti-Crk (C-terminal region) (lanes 5 & 6).

Background: Crk

Crk belongs to a family of adaptor molecules, which link extracellular growth and differentiation stimuli with formation of signal transduction protein complexes. Crk is mainly composed of Src homology 2 (SH2) and Src homology 3 (SH3) domains, which serve as docking sites for signalling proteins, including various receptors, cytoplasmic kinases, and GTPase regulators. These signal transduction pathways lead to modulation of cell adhesion, cell migration and immune cell responses.
