Immunohistochemistry-Paraffin: CXCR3 Antibody [NB100-56404] - IHC analysis of a FFPE human spleen using 1:25 dilution of CXCR3 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of HIER using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (40X) was performed using Aperio AT2 (Leica Biosystems). Staining was performed by Histowiz.

Background: CXCR3

Chemokines play important roles in inflammation and critical for the recruitment of effector immune cells to sites of infection. Historically, CD183 was the third CXC chemokine receptor discovered and therefore, commonly designated as CXCR3. Binding of chemokines to CD183 induces cellular responses that are involved in leukocyte traffic, most notably integrin activation, cytoskeletal changes and chemotactic migration. The CXC chemokine receptor (CXCR) 3, a G-protein-coupled 7-transmembrane receptor, is expressed at high levels on activated and memory T cells, B cells, natural killer cells, and plasmacytoid monocytes.
