Western Blot: DDIT4 Antibody (PS00-10) [NBP3-32259] - Western blot analysis of DDIT4 on different lysates with Rabbit anti-DDIT4 antibody (NBP3-32259) at 1/1,000 dilution.

Lane 1: HeLa cell lysate
Lane 2: HeLa treated with 0.5mM CoCl2 for 6 hours cell lysate

Lysates/proteins at 20 ug/Lane.

Predicted band size: 25 kDa
Observed band size: 30 kDa

Exposure time: 50 seconds;

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32259) at 1/1,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Background: DDIT4

REDD1 (regulated in development and DNA damage response 1) is a direct transcriptional target of HIF. It plays a role in the TSC/mTOR pathway as a regulator upstream of the TSC1/2 complex. In response to both hypoxia and energy stress, REDD1 inhibits mTOR activity in a TSC-dependent manner.
