Western Blot: Desmoglein-3 Antibody (5H10) - Azide and BSA Free [NBP2-80695] - Colocalisation of Dsg3 with E-cadherin-catenin complex. Co-IPs demonstrated that both endogenous and exogenous Dsg3 interacted with the E-cadherin-catenin complex in A431 stable lines (the empty vector control (V) and hDsg3.myc (D3)) as well as in HaCaTs. When pulled down with antibodies against the myc tag or with a mouse Dsg3 Ab 5H10 and blotted for E-cadherin and beta-catenin, both proteins were detected in D3 and, to a lesser extent, in V. Conversely, when pulled down with E-cadherin Ab HECD-1 and blotted for Dsg3 or the myc tag and Pg, these proteins were detected in both V and D3 cells. Note that this result also was confirmed in HaCaTs (HaC) where Dsg3 was expressed endogenously (far right blot). Background: Desmoglein-3

Desmoglein 3 (Dsg3) is a member of sub-family the desmoglein (DSGs) and it mainly serves as adhesion component of intercellular desmosome junctions. DSGs serve essential part of the core complex of desmosomes, and together with desmocollin (DSCs), they represents transmembrane proteins which mediate adhesion by transinteracting with cadherins of the opposing cell. Homophilic (e.g., DSG3-DSG3) as well as heterophilic (e.g., DSC3-DSG1) transinteraction have been reported for these proteins and within the desmosome, DSGs/DSCs are anchored to the intracellular plaque proteins plakoglobin, plakophilins, and desmoplakin, the latter of which mediates connection to the intermediate filament cytoskeleton. Desmoglein 3 is predominately expressed in stratified squamous epithelia including epidermis, tongue, tonsil, esophagus and carcinomas. Desmoglein 3 has been characterized as an auto-antigen of pemphigus vulgaris (PV), a life-threatening autoimmune disease, wherein the binding of anti-desmoglein 3 autoantibodies causes the loss of cell-cell adhesion, which clinically manifests as blisters on the skin and oral mucosa, and histologically as suprabasilar acantholysis.
