Lane 1: MCF7 cell lysate
Lane 2: MCF7 serum starved for 6 hours then add 10nM Neuregulin-1 for 10 minutes cell lysate
Lane 3: MCF7 serum starved for 6 hours then add 10nM Neuregulin-1 for 10 minutes cell lysate, then the membrane treated with pp for 1 hour

Lysates/proteins at 20 ug/Lane.

Predicted band size: 148 kDa
Observed band size: 180 kDa

Exposure time: 3 minutes; ECL;

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32765) at 1/1,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/ Immunofluorescence: ErbB3/Her3 [p Tyr1289] Antibody (JE51-25) [NBP3-32765] - Immunocytochemistry analysis of MCF7 cells serum starved for 6 hours then add 10nM Neuregulin-1 for 10 minutes labeling ErbB3/Her3 with Rabbit anti-ErbB3/Her3 antibody (NBP3-32765) at 1/100 dilution.

Cells were fixed in 4% paraformaldehyde for 20 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 5 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature. Cells were then incubated with Rabbit anti-ErbB3/Her3 antibody (NBP3-32765) at 1/100 dilution in 1% BSA in PBST overnight at 4 ℃. Goat Anti-Rabbit IgG H&L (iFluor™ 488) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. Nuclear DNA was labelled in blue with DAPI.

Beta tubulin (red) was stained at 1/100 dilution overnight at +4℃. Goat Anti-Mouse IgG H&L (iFluor™ 594) was used as the secondary antibody at 1/1,000 dilution.

Background: ErbB3/Her3

The ErbB3 gene encodes a member of the epidermal growth factor receptor (EGFR) family of receptor tyrosine kinases. ErbB3 is a membrane-bound protein which has a neuregulin binding domain but not an active kinase domain. It can therefore bind this ligand but cannot convey a signal into the cell via protein phosphorylation. However it does form heterodimers with other EGF receptor family members which do have kinase activity. Heterodimerization leads to the activation of pathways which lead to cell proliferation or differentiation. Amplification of this gene and/or overexpression of its protein have been reported in numerous cancers including prostate, bladder and breast tumors. Alternate transcriptional splice variants encoding different isoforms have been characterized. One isoform lacks the intermembrane region and is secreted outside the cell. This form acts to modulate the activity of the membrane-bound form. Additional splice variants have also been reported but they have not been thoroughly characterized.
