A synthetic peptide corresponding to amino acids 75-92 (DRKAVETHLLRNPHLVS) of human FLIP was used as immunogen, GenBank no. gi|12643547. This amino acid sequnce is 100% conserved in FLIP long and short, and various FLIP isoforms as denoted in the NCBI BLAST data base. http://www.ncbi.nlm.nih.gov/blast/.

Specificity

The gene encoding FLIP produces, by alternative splicing, 24 different transcripts, all with introns, putatively encoding 22 different protein isoforms.

Background: FLIP

Cell death signals are transduced by death domain (DD)- containing adapter molecules and members of the caspase (ICE/CED-3) protease family. Most if not all, forms of apoptosis involve activation of the caspases. Caspases-8 (FLICE) and -10 (FLICE2) are two pivotal members in the caspase family. FLICE-inhibitory proteins were identified in virus and human and designated v-FLIPs and FLIP, respectively. The human FLIP was also cloned by several labs independently and termed Casper , I-FLICE, FLAME-1, CASH and CLARP3-7. FLIP contains two death effector domains (DEDs) and a caspase-like domain. FLIP interacts with adapter protein FADD and caspase-8 and 10, and potently inhibits apoptosis induced by death receptors (reviewed in Peter, 2004). However, the functions of FLIP remain to be fully elucidated and there is some evidence FLIP may also act as an activator of caspase-8/-10 in certain cases, thereby taking on an apoptosis promoting role. FLIP has a short form and a long form. Additionally, four splice variants of c-FLIPs have been identified and termed FLIP a, b, g, and d. NB100-56141 recognizes FLIP. Human FLIP long is a 480 amino acid protein, GenBank no. gi|12643547. Human FLIP short is a 221 amino acid protein, GenBank no. AAC51623.1. NB100-56141 will recognize FLIP long and short, and FLIP isoforms that contain the peptide immunogen sequence (DRKAVETHLLRNPHLVSD), including FLIP a, b, g, and d.
