Knockout Validated: FUS Antibody (CL0190) [NBP2-52874] - Immunofluorescence of HeLa wildtype (WT) and FUS knockout (KO) cells labelled with a green or a far-red fluorescent dye, respectively. WT and KO cells were mixed and plated to a 1:1 ratio on coverslips. Cells were stained with Mouse Anti-FUS Monoclonal Antibody (Catalog #NBP2-52874) at 1:1000, followed by the corresponding Alexa Fluor 555-coupled Secondary Antibody. Acquisition of the green (identification of WT cells), red (antibody staining) and far-red (identification of KO cells) channels was performed. Knockout Validated: FUS Antibody (CL0190) [NBP2-52874] - Western blot using lysates of HeLa parental cell line (WT) and FUS knockout HeLa cell line (KO), collected in RIPA Buffer. Lysates were prepared using 30 ug of protein and the nitrocellulose membrane was probed with Mouse Anti-FUS Monoclonal Antibody (Catalog #NBP2-52874) at 1:5000 O/N at 4C, followed by peroxidase-conjugated Anti-Mouse Secondary Antibody and ECL detection. A specific band at ~70 kDa was detected in the FUS WT cell line but is not observed in the FUS KO cell line. The Ponceau stained transfer of the blot is shown to confirm equal protein loading. Background: FUS

FUS, fused in sarcoma, encodes a multifunctional protein component of the heterogeneous nuclear ribonucleoprotein (hnRNP) complex. Nuclear receptor coactivator that directly binds nuclear receptors and stimulates the transcriptional activities in a hormone-dependent fashion. Coactivates expression in an agonist- and AF2-dependent manner. Involved in the coactivation of different nuclear receptors, such as for steroids (GR and ERs), retinoids (RARs and RXRs), thyroid hormone (TRs), vitamin D3 (VDR) and prostanoids (PPARs). Probably functions as a general coactivator, rather than just a nuclear receptor coactivator. May also be involved in the coactivation of the NF-kappa-B pathway. May coactivate expression via a remodeling of chromatin and its interaction with histone acetyltransferase proteins. Involved in placental, cardiac, hepatic and embryonic development. NCOA6 is over-expressed in several breast cancer cell lines.
