Western Blot: Galectin-3 Antibody (5C21) [NBP1-92690]

Western Blot: Galectin-3 Antibody (5C21) [NBP1-92690] - Analysis of different tissue and cell lysates using mouse mAb to galectin-3, NBP1-92690, dilution 1:2,000 in green: [1] protein standard (red), [2] mouse brain lysate, rat tissue lysates: [3] brain, [4] liver, [5] heart, [6] kidney, [7] lung, and [8] HEK293 cell lysate. The band at about 30kDa mark corresponds to the galectin-3 protein. The same blot was simultaneously probed with chicken pAb to HSP60, dilution 1:20,000, in red, which reveals HSP60 protein in all preparations.

Immunocytochemistry/Immunofluorescence: Galectin-3 Antibody (5C21) [NBP1-92690] - Analysis of cortical neuron-glial cell culture from E20 rat stained with mouse mAb to galectin-3, NBP1-92690, dilution 1:1,000 in red, and costained with chicken pAb to MAP2, dilution 1:10,000 in green. The blue is Hoechst staining of nuclear DNA. The NBP1-92690 antibody produces strong staining of microglia (top panel), other glial cells (bottom panel) and lymphocytic cells (small round cells), while the MAP2 antibody labels dendrites and perikarya of mature neurons.

Background: Galectin-3

Galectin-3 (LGALS3 or Mac-2 antigen), is a 26 kD beta-galactoside-binding protein from galectin family and it is ubiquitously expressed with localization dependent on cell type as well as proliferative status. Galectin-3 is expressed in many immunocompetent and inflammatory cells, including macrophages, dendritic cells, eosinophils, mast cells, uterine NK cells and activated T and B cells, and it is localized in nucleus, cytoplasm or on cell surface as well as in extracellular compartments. Extracellularly, galectin-3 acts as an adhesion molecule whereas intracellular galectin-3 implicates in processes that are pivotal for basic cellular functions, such as pre-mRNA splicing regulation of cell growth, cell cycle progression and apoptosis. Galectin-3 binds and interacts with numerous ligands in intra-/extracellular environment such as DMBT1, ITGA3, ITGB1, CSPG4, LGALS3BP, LYPD3, CYHR1, UACA etc. for modulating cellular signals responsible fpr its biological functions such as phagocytosis, cell growth/proliferation, adhesion and apoptosis in normal as well as cancer cells. Circulating galectin-3 concentrations increases more than 30-fold in bloodstream of patients with various cancers and patients with metastatic disease have higher concentrations of circulating galectin-3 than those with only localized tumors.
