Immunocytochemistry/Immunofluorescence: GR/NR3C1 Antibody (59) [NB120-3579] - Analysis of Glucocorticoid Receptor using Glucocorticoid Receptor Polyclonal Antibody (59) shows staining in Hela Cells. Glucocorticoid Receptor (green), F-Actin staining with Phalloidin (red) and nuclei with DAPI (blue) is shown. Cells were grown on chamber slides and fixed with formaldehyde prior to staining. Cells were probed without (control) or with an antibody recognizing Glucocorticoid Receptor at a dilution of 1:100 over night at 4C, washed with PBS and incubated with a DyLight-488 conjugated.

Application Notes
WB: Detects an approx. 97 kDa protein representing GR as well as two other unidentified proteins at approx. 135 - 145 kDa from rat liver extract. IHC: Staining of GR in rat hippocampus using NB120-3579 results in staining primarily in the nucleus. NB120-3579 does not disrupt receptor-DNA interactions.

Background: GR/NR3C1

Steroid receptors are ligand-dependent, intracellular proteins that stimulate transcription of specific genes by binding to specific DNA sequences following activation by the appropriate hormone. Glucocorticoids are a family of steroids necessary for the regulation of energy metabolism and the immune and inflammatory responses. These compounds exert their effect through their interaction with the glucocoticoid receptor (GR) and that complex's subsequent association with DNA. All normal mammalian tissues examined to date have been shown to contain glucocorticoid receptor. The corresponding gene for the glucocorticoid receptor is NR3C1.
