After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4C overnight. Immunocytochemistry/Immunofluorescence: Granzyme B Antibody (6A1) [NBP3-26241] - Staining of Hela Cells with Granzyme B Antibody (6A1) at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeated by 0.2% Triton X-100, and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. Nuclear DNA was labeled in blue with DAPI. The secondary antibody was FITC-conjugated Goat Anti-Rabbit IgG (H+L).Applications for Granzyme B Antibody (6A1)

Background: Granzyme B
Granzyme family serine proteases are stored in the granules of cytotoxic T lymphocytes and natural killer cells. Granzymes contain one S1 protease domain and are synthesized as preproproteins. Granzymes are released toward pathogen infected or transformed target cells during cellular immune reactions. Target cell entry by granzymes is through perforin channels. Granzymes trigger apoptosis by caspase-dependent and -independent mechanisms. There are five granzymes (A, B, G, H and K) in human and many more in mouse and rat.
