Immunocytochemistry/Immunofluorescence: GRK1 Antibody (D11) [NB120-2776] - Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes at room temperature and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with a Rhodopsin Kinase 1a/b Monclonal Antibody (D11) at a dilution of 1:200 and incubated overnight in a humidified chamber. Cells were washed with PBST and incubated with a DyLight-conjugated secondary antibody for 45 minutes at room temperature in the dark. Immunocytochemistry/Immunofluorescence: GRK1 Antibody (D11) [NB120-2776] - Analysis of Rhodopsin Kinase 1a/b (green) in Hela cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes at room temperature and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with a Rhodopsin Kinase 1a/b Monclonal Antibody (D11) at a dilution of 1:200 and incubated overnight in a humidified chamber. Cells were washed with PBST and incubated with a DyLight-conjugated secondary antibody for 45 minutes at room temperature in the dark. WB: Detects an approx. 60 kDa protein representing GRK 1a/b. ab2766 detects both in vitro expressed splice variants by WB but was not able to detect significant amounts of GRK1b from human retina. IF: Staining of GRK1a/b in bovine retina with ab2766 results in intense staining primarily in cone outer segments, but also in rod outer segments. Weak staining is observed in somata and synaptic terminals of cones and inner segments of rods.
