Specificity

This monoclonal antibody reacts with cells bearing HLA-A25 or HLA-Aw32 antigens. In addition, a reaction was observed with a cell of phenotype A2, Aw31; B17, Bw49. HLA-A, with HLA-B and HLA-C, belongs to major histocompatibility complex (MHC) class I antigens and expresses constitutively on all nucleated cells. HLA system comprises closely linked genes controlling highly polymorphic proteins involved in the presentation of peptides to the T-cell receptor, inhibition of NK cell cytotoxicity, and rejection of tissue allotransplantation. Specific alleles at HLA loci are associated with diseases. This monoclonal antibody is specifically applicable for typing peripheral T cells for the antigens HLA-A25 and HLA-Aw32.

Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.

Background: HLA A

HLA-A belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exons 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region, and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. Typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. Hundreds of HLA-A alleles have been described. [provided by RefSeq]
