Immunoprecipitation: HMGN3/TRIP7 Antibody [NB100-59839] - Detection of human HMGN3 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-HMGN3 antibody NB100-59839 (lot 2) used for IP at 6 ug per reaction. HMGN3 was also immunoprecipitated with a previous lot of this antibody (lot 1) and rabbit anti-HMGN3 antibody NB100-59840 (lots 1 and 2). For blotting immunoprecipitated HGMN3, NB100-59840 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background: HMGN3/TRIP7

HMGN3 was originally identified in a screen for interaction partners of the thyroid hormone receptor. HMGN3 belongs to the HMGN family of small basic proteins that interacts specifically with nucleosomes and functions to facilitate chromatin unfolding to allow access of transcription factors to DNA targets. HMGN has been found to be a regulator of the glycine transporter 1 (GLYT1) gene.
