Background: HSP70/HSPA1A

The 70 kilodalton heat shock proteins (Hsp70s) are a family of ubiquitously expressed proteins. All of the Hsp70 proteins have three major functional domains. An N-terminal ATPase domain binds ATP (Adenosine triphosphate) and hydrolyzes it to ADP (Adenosine diphosphate), a substrate binding domain contains a groove with an affinity for neutral, hydrophobic amino acid residues and a C-terminal domain rich in alpha helical structure acts as a 'lid' for the substrate binding domain. Eukaryotic organisms express several slightly different Hsp70 proteins. Hsc70 or Hsp73; a constitutively expressed chaperone protein, Hsp70 or Hsp72; a stress-induced protein, BiP or Grp78; a protein localized to the endoplasmic reticulum mtHsp70 or Grp75; the mitochondrial Hsp70. Heat shock protein 72 (Hsp72) is the major inducible, 70kDa heat shock protein. It plays a role in many cellular activities including protein synthesis, folding, and translocation into organelles as well as the assembly of multiprotein complexes. Hsc70 typically makes up one to three percent of total cellular protein. The genes are located in the major histocompatibility complex class III region.
