Detection of Human CD74 by Western Blot.
Western blot shows lysates of Raji human Burkitt's lymphoma cell line and Daudi human Burkitt's lymphoma cell line. PVDF Membrane was probed with 1 µg/mL of Human CD74 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3590) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for CD74 at approximately 35 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.Detection of Human CD74 by Simple WesternTM.
Simple Western lane view shows lysates of Raji human Burkitt's lymphoma cell line and Daudi human Burkitt's lymphoma cell line, loaded at 0.2 mg/mL. Specific bands were detected for CD74 at approximately 49 & 50 kDa (as indicated) using 10 µg/mL of Sheep Anti-Human CD74 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3590) followed by 1:50 dilution of HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). Background: CD74
CD74 is a transmembrane protein which, on antigen presenting cells, is the invariant chain that chaperones MHC class II dimers from the endoplasmic reticulum to the cell surface. The cytoplasmic portion of CD74 can be released by intramembrane cleavage and activate NF kappaB. CD74 is upregulated in several cancers and is also expressed by non-immune cells during inflammation. This antibody was raised to a splice variant of human CD74 which lacks the Thyroglobulin type-1 repeat in the extracellular domain. Within comparable regions, human CD74 shares 70% and 73% amino acid sequence identity with mouse and rat CD74, respectively.
