Detection of Human Chondroadherin by Western Blot.
Western blot shows lysates of human cartilage tissue. PVDF membrane was probed with 2 µg/mL of Sheep Anti-Human Chondroadherin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8218) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Chondroadherin at approximately 34-36 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Chondroadherin in Human Cartilage.
Chondroadherin was detected in immersion fixed paraffin-embedded sections of human cartilage using Sheep Anti-Human Chondroadherin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF8218) at 1 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). Background: Chondroadherin
Chondroadherin is an approximately 36 kDa secreted glycosaminoglycan that contains nine tandem leucine-rich repeats as well as N-terminal and C-terminal leucine-rich domains. It is a component of the extracellular matrix in cartilage, tendon, bony growth plates, skeletal and cardiac muscle, lung, gastrointestinal tract, pancreas, cerebellum, and eye. It is cleaved by HTRA1/Omi during intervertebral disc degeneration. Chondroadherin functions as an adhesion protein by binding to Integrin a2/b1, Collagen II, and Collagen VI. It also promotes complement activation through binding to C1q and Complement Factor H. Mature human Chondroadherin shares 94% amino acid sequence identity with mouse and rat Chondroadherin.
