Specificity

Detects human IL-17/IL-17A in direct ELISAs. In direct ELISAs, approximately 40% cross-reactivity with recombinant human (rh) IL-17A/IL-17F heterodimer is observed. No cross-reactivity with recombinant mouse IL-17/IL-17A, recombinant canine IL-17/IL-17A, rhIL-17B, rhIL-17C, rhIL-17D, rhIL-17E, or rhIL-17F is observed.

Human peripheral blood mononuclear cells (PBMCs) treated with PMA, Calcium Ionomycin, LPS, and Recombinant Human IL-23 (Catalog # 1290-IL) to induce Th17 development overnight, re-stimulated with PMA, Calcium Ionomycin, and Monensin for 2 hours were stained with Mouse Anti-Human IL-17/IL-17A Alexa Fluor® 488-conjugated Monoclonal Antibody (Catalog # IC3171G, filled histogram) or isotype control antibody (Catalog # IC002G, open histogram). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005).
