Detection of IL‑9 in Human PBMCs by Flow Cytometry.

Human peripheral blood mononuclear cells (PBMCs) were treated with plate-bound anti-CD3 Monoclonal Antibody (5 μg/ml) and anti-CD28 Monoclonal Antibody (2 μg/ml), Recombinant Human IL-4 (204-IL,10 ng/ml), and anti-Human IFN-gamma Monoclonal Antibody (AF-285-NA, 20 μg/ml) for 5 days and stained with (A) Mouse Anti-Human IL-9 APC-conjugated Monoclonal Antibody (Catalog # IC209A) or (B) Mouse IgG2B Isotype Control Antibody (IC0041A) and Mouse Anti-Human IFN-gamma Phycoerythrin-conjugated Monoclonal Antibody (IC285P). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (FC005). Staining was performed using our Staining Intracellular Molecules protocol.

Background: IL-9

Interleukin-9 (IL-9) is a 28-30 kDa monomeric glycosylated polypeptide that belongs to the IL-7/IL-9 family of proteins. Although multiple cell types are reported to express IL-9, including Th17 cells, eosinophils, basophils, NK T cells and mast cells, the two cell types possibly most central to the process are Th9 and ILC-2 (or innate lymphoid cell type 2) cells. IL-9 supports the growth of multiple hematopoietic cell types including germinal center B cells, macrophages, mast cells, neutrophils, megakaryocytes, and erythrocytes. It also promotes IgE production, the activation and chemoattraction of mast cells, mucus production by epithelium, Th17 cell proliferation and TGF-beta production by dendritic cells. IL-9 exerts its biological effects through a receptor complex composed of IL-9 R and the common gamma chain. Mature human IL-9 shares 57% amino acid sequence identity with mouse and rat IL-9.
