Detection of Human ITIH2 by Western Blot.

Western blot shows lysates of HepG2 human hepatocellular carcinoma cell line. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human ITIH2 Monoclonal Antibody (Catalog # MAB101991) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (HAF018). A specific band was detected for ITIH2 at approximately 106 kDa (as indicated). This experiment was conducted under reducing conditions and using Western Blot Buffer Group 1.

Human ITIH2 ELISA Standard Curve.

Recombinant Human ITIH2 protein was serially diluted 2-fold and captured by Mouse Anti-Human ITIH2 Monoclonal Antibody (MAB101993) coated on a Clear Polystyrene Microplate (DY990). Mouse Anti-Human ITIH2 Monoclonal Antibody (Catalog # MAB101991) was biotinylated and incubated with the protein captured on the plate. Detection of the standard curve was achieved by incubating Streptavidin-HRP (DY998) followed by Substrate Solution (DY999) and stopping the enzymatic reaction with Stop Solution (DY994).

Background: ITIH2

The ITIs are a family of plasma protease inhibitors contributing to extracellular matrix stabilization by covalent linkage to hyaluronan.  ITIH2 is a binding protein between hyaluronan and other matrix proteins.  It regulates the localization, synthesis and degradation of hyaluronan.  ITIH molecules are shown to play a role in carcinogenesis and inflammation.  ITIH genes are downregulated in multiple human solid tumors.  This downregulation contributes to initiation and/or progression of malignancy.
