Detection of P2X7/P2RX7 in Lung Cancer.
P2X7/P2RX7 was detected in immersion fixed paraffin-embedded sections of Lung Cancer using Mouse Anti-Human P2X7/P2RX7 Monoclonal Antibody (Catalog # MAB97902) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using VisUCyte Antigen Retrieval Reagent-Basic (Catalog # VCTS021). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cell surface in cancer cells. Background: P2X7/P2RX7
Human P2X purinoceptor 7 (aka P2X7) is a 595 aminoacids (aa) protein encoded by the P2RX7 gene. P2X7 receptors belong to the family of ATP-gated ion channels, and their activity can be found in cells of hemopoietic lineage including macrophages, microglia, and certain lymphocytes. They mediate the influx of Ca2+ and Na+ and the release of pro-inflammatory cytokines. At least 8 different human P2X7 isoforms formed by alternative splicing have been reported, ranging from 128 aa to 595 aa. The P2X7 subunits can form homomeric receptors only with a typical P2X receptor structure.
