Detection of Phospho-IFN-alpha / beta R1 (S535/S539) by Western Blot

Western blot of immunoprecipitates from HEK293 cells transfected with 1) Mock, 2) IFN-alpha R1 wild type, and 3) IFN-alpha R1 S535A and S539A mutants showing specific immunolabeling of the ~110 kDa to ~130 kDa IFN-alpha R1 wild type. The immunolabeling is absent in the IFN-alpha R1 S535 and S539 mutants (Control). The immunolableing is blocked by the phosphopeptide (Phos) used as the antigen but not by the corresponding dephosphopeptide (Dephos).

The molecular weight of the IFN-alpha R1 varies with cell line (different levels of glycosylation) in HEK293 and HeLa cells; the mature form is ~110 - 130 kDa.

Background: IFN-alpha/beta R1

Interferons are widely used therapeutic agents because of their anti tumor and antiviral effects and because of their modulatory effects on the immune system (1,2). These cytokines produce their effects by binding to the Type I IFN Receptor. This receptor contains two subunits; IFNAR1/ alpha1 and IFNAR2/ alpha2 Down regulation of this receptor plays a key role in determining the magnitude and duration of cytokine signaling. This downregulation is thought to be influenced by phosphorylation of S535 and S539 in IFN-alpha R1 (3).
