Human peripheral blood mononuclear cells (PBMCs) either (A) untreated or (B) treated with PMA, Calcium Ionomycin, LPS, Recombinant Human IL-6 (Catalog # 206-IL) and Recombinant Human IL-23 (Catalog # 1290-IL) were stained with Mouse Anti-Human ROR gamma/RORC/NR1F3 PerCP-conjugated Monoclonal Antibody (Catalog # IC6109C) and Mouse Anti-Human CD3e APC-conjugated Monoclonal Antibody (Catalog # FAB100A). Quadrant markers were set based on control antibody staining (Catalog # IC0041C). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Background: ROR gamma/RORC/NR1F3
ROR gamma (Nuclear receptor ROR-gamma), also called RORC and NR1F3, is a member of the NR1 nuclear hormone receptor family. ROR gamma is a DNA binding transcription factor. ROR gamma is 518 amino acids (aa) in length. Deletion in mice implicates ROR gamma as being essential for lymphoid organogenesis and controlling apoptosis during thymopoiesis. Two splice forms differing in the first 24 aa have been found for this gene; isoform 2 deletes aa 1‑21 resulting in an alteration of aa 22‑24. Over aa 1‑100 human RORC shares 96% identity with mouse ROR gamma.
