S100P in Human Stomach.
S100P was detected in immersion fixed paraffin-embedded sections of human stomach using Mouse Anti-Human S100P Monoclonal Antibody (Catalog # MAB29571) at 0.5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in gastric glands. Background: S100P
Human S100P is a 22 kDa, homodimeric member of the S100 family of calcium-binding proteins. The S100 family currently has more than 20 members and belongs to the EF-hand superfamily of molecules. S100P is 95 aa in length and contains short, sequential modules. There is an N-terminal alpha-helix, a unique EF-hand motif, an alpha-helix, a linker region, an alpha-helix, a classic EF-hand motif and a C-terminal alpha-helix. The EF-hand motif binds calcium, which likely alters molecular conformation. The rearranged S100P now binds ligand with the linker region. Human S100P shares 47% aa sequence identity with rat S100P.
