Immunoprecipitation: KLHL12/C3IP1 Antibody [NBP1-49927] - Detection of Human KLHL12 by Western Blot of Immunoprecipitates. Samples: Whole cell lysate (1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-KLHL12 antibody used for IP at 3 mcg/mg lysate. KLHL12 was also immunoprecipitated by rabbit anti-KLHL12 antibody which recognizes a downstream epitope. For blotting immunoprecipitated KLHL12 was used at 1 mcg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Antibodies: Affinity purified rabbit anti-KLHL12 antibody NBP1-49927 (lot NBP1-49927-2 ) used for IP at 6 ug per reaction. KLHL12 was also immunoprecipitated by a previous lot of this antibody NBP1-49927-1. For blotting immunoprecipitated KLHL12, NBP1-49927 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background: KLHL12/C3IP1

Serves as a substrate-specific adapter for the CUL3-based ubiquitin-protein E3 ligase complex. Negatively regulates the Wnt signaling pathway via the targeted ubiquitination and subsequent proteolysis of DVL3. Component of an ubiquitin-protein E3 ligase complex which includes at least CUL3 and KLHL12. This complex interacts with DVL3 upon activation of the Wnt signaling pathway by WNT3A.
