Specificity

Western Blot: MAGEA3 Antibody (21B4) - Chimeric - Azide and BSA Free [NBP3-20167] - HeLa(A) (0.01ug/ml) cells lysate and HeLa(B) (0.01ug/ml) cells nuclear lysate (35ug protein in RIPA buffer) were resolved on a SDS PAGE gel and blots were probed with the chimeric rabbit version of 21B4 (NBP3-20167) before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence.

Immunocytochemistry/Immunofluorescence: MAGEA3 Antibody (21B4) - Chimeric - Azide and BSA Free [NBP3-20167] - Immunofluorescence analysis of paraformaldehyde fixed HeLa cells stained with the chimeric rabbit IgG version of 21B4 (NBP3-20167) (1:1000 dilution) for 1h followed by Alexa Fluor(R) 488 secondary antibody (1:1500 dilution), showing nuclear staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-20167, DAPI, merged channels and an isotype control. The isotype control was anti-fluorescein antibody (NBP2-52638) followed by staining with Alexa Fluor(R) 488 secondary antibody.

Flow Cytometry: MAGEA3 Antibody (21B4) - Chimeric - Azide and BSA Free [NBP3-20167] - HeLa cells were stained with an isotype control (NBP2-52638, black line) or the rabbit-chimeric version of 21B4 (NBP3-20167, blue line) at a concentration of 1 ug/ml for 30 mins at RT. After washing, bound antibody was detected using Alexa Fluor 488(R) conjugated goat anti-rabbit IgG and cells analysed on a FACSCanto flow-cytometer.

Background: MAGEA3

MAGEA3, also known as Melanoma-associated antigen 3, is a 34.7 kDa, 314 amino acid protein that is involved in increasing ubiquitin ligase activity as a member of the MAGEA family. Current research is being conducted to study the effects of the protein on diseases and disorders such as bile duct cancer, dyskeratosis, ocular melanoma, germ cell tumors, multiple types of carcinomas, avian influenza, testicular cancer, lung cancer, and lymphoma. The protein interacts with other proteins such as TRIM28, UBC, HTT, and SUMO2.
