Immunogen

This antibody was raised by immunising Lewis 1F rats with mouse CH-1 lymphoma cells. Spleen cells from immunised rats were fused with X-63Ag8-653 myeloma cells to generate hybridomas.

Specificity

This antibody is specific for murine MHC class I, and binds to polymorphic H-2 determinants expressed on most haplotypes. This antibody is, therefore, able to bind all H-2 class I molecules, independent of the allele. This antibody recognises a determinant distinct from those detected by most common mouse monoclonal anti-H-2 antibodies.

Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original format, for improved compatibility with existing reagents, assays and techniques.

This antibody binds murine H-2 MHC class I molecules, and has been used in both structural and biochemical studies. This antibody has been used in immunoprecipitation analysis (Robinson, 1987), and in immunofluorescence assays for MHC-I surface expression (Karacsonyi et al, 2004) and MHC-I endocytosis (Knorr et al, 2009).This antibody has also been employed to induce clustering of cell surface MHC-I in co-patching assays (Knorr et al, 2009). Cellular radioimmune assays have been used to confirm the binding of this antibody to spleen cells, and assess its strain specificity (Koch et al, 1983).

Background: MHC Class I

The antigenic agglomerate major histocompatibility complex (MHC) class I molecules present antigenic peptides to CD8-positive T cells. It is now recognized that the products of the MHC genes are antigen-presenting molecules (APM) designed for the presentation of antigen fragments (peptides) to the T-cell receptor, thus participating in the immune response (1). Recently shown is a mechanism of antigen acquisition for cross-presentation that couples the antigen presentation system of two adjacent cells and is lost in most tumours: gap-junction-mediated intercellular peptide coupling for presentation by bystander MHC class I molecules and transfer to professional antigen presenting cells for cross-priming (2). The mammalian vomeronasal organ detects social information about gender, status, and individuality. It has been shown that small peptides that serve as ligands for major histocompatibility complex (MHC) class I molecules function also as sensory stimuli for a subset of vomeronasal sensory neurons located in the basal Gao- and V2R receptor-expressing zone of the vomeronasal epithelium (3).
