Integrin alpha 9 in Mouse Liver Tissue.

Integrin a9 was detected in perfusion fixed frozen sections of mouse liver tissue using Goat Anti-Mouse Integrin a9 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF3827) at 1 µg/mL overnight at 4 °C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using the Anti-Goat IgG VisUCyte™ HRP Polymer Antibody (brown; Catalog # VC004) and counterstained with hematoxylin (blue). Specific staining was localized to bile canaliculi. Intergins alpha4/ alpha9 are responsible for the action of osteopontin (OPN) in the augmentation of macrophage (Mφ) M2 polarization and angiogenic capacity. (A) Relative mean fluorescence intensities (MFIs) of integrins alpha4/ alpha9 were measured by FACS analysis in each Mφ subset. Anti-integrin alpha4 or alpha9 antibodies (Abs) and each isotype-matched control Ab were used at 10 µg/mL, n = 4 [***p < 0.001 vs. untreated, ###p < 0.001 vs. interleukin (IL)-10 alone]. Background: Integrin alpha 9

Integrin alpha9 is a 150 kDa type I transmembrane glycoprotein that is widely expressed and is found on smooth muscle, keratinocytes, skeletal muscle and hepatocytes. It principally associates with the beta1 integrin. alpha9 beta1 binds to VEGF-C, VEGF-D, and osteopontin. The 951 amino acid (aa) extracellular domain of mouse integrin alpha9 contains three beta-propellor repeats and multiple PheGly-GlyAlaPro repeats. A potential proteolytic cleavage site in human alpha9 ECD is absolutely conserved in mouse alpha9 ECD (Arg566-Val567). The ECD of mouse integrin alpha9 shares 95% and 89% aa sequence identity to rat and human integrin alpha9, respectively.
