Recombinant Mouse R-Spondin 1 (7150-RS) co-stimulates beta-catenin response in the HEK293T human embryonic kidney cell line in a dose-dependent manner (orange line). beta-Catenin Response elicited by Recombinant Mouse R-Spondin 1 (500 ng/mL) is neutralized (green line) by increasing concentrations of Rat Anti-Mouse R-Spondin 1 Mono-clonal Antibody (Catalog # MAB3474) in the presence of 5 ng/mL of Recombinant Mouse Wnt-3a (1324-WN). The ND50 is typically 0.4-2 µg/mL.

Neutralization

Measured by its ability to neutralize R‑Spondin 1-induced beta-catenin response in the HEK293T human embryonic kidney cell line. The Neutralization Dose (ND50) is typically 0.4-2 μg/mL in the presence of 500 ng/mL Recombinant Mouse R‑Spondin 1 and 5 ng/mL Recombinant Mouse Wnt-3a.

Background: R-Spondin 1

RSPO-1 (roof plate-specific spondin; also R-Spondin 1 and cristin3) is a 39 kDa, heparin-binding member of the Cristin/R-Spondin family of proteins. These molecules apparently are either retained intracellularly or secreted, where they activate the frizzled-LRP receptor pathway. Mature human RSPO-1 is 27 kDa and 243 amino acids (aa) in length. It contains a cysteine-rich furin repeat domain followed by a type I TSP domain. This form is apparently retained intracellularly. A presumably glycosylated, 30 kDa, 193 aa C-terminally truncated form is secreted and interacts with heparin. The secreted form of human RSPO-1 is 93% aa identical to secreted mouse RSPO-1.
