Detection of Mouse RTVP-1 by Western Blot.
Western blot shows lysates of Balb/3T3 mouse embryonic fibroblast cell line and RAW 264.7 mouse monocyte/macrophage cell line. PVDF membrane was probed with 1 µg/mL of Mouse RTVP-1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4468) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for RTVP-1 at approximately 30 kDa (as indicated). Background: RTVP-1
RTVP-1 (related to testes-specific, vespid, and pathogenesis protein 1) can act as both a tumor suppressor and tumor promoter gene. RTVP-1 is a tumor suppressor gene based on its regulation by p53, induction of apoptosis, growth inhibition, and decreased expression in prostate tumors when compared to normal prostate. RTVP-1 is a tumor promoter is based on evidence that overexpression of RTVP-1 decreased the phosphorylation of c-Jun, increased Bcl-2 expression, and increased MMP2 activity. In addition, RTVP-1 is expressed at high levels in glioblastomas while barely detectable in normal brain/astrocytes. Differences in the levels of cellular and extracellular forms as well as different cellular processing of RTVP-1 may account for these differing roles. Additionally, increased RTVP-1 expression is associated with myelomocytic differentiation in macrophages.
