Immunoprecipitation:Myosin Heavy Chain AntibodyNBP3-29780] -Detection of human MYH14 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-MYH14 antibody NBP3-29780 (lot NBP3-29780-1) used for IP at 6 µg per reaction. MYH14 was also immunoprecipitated by rabbit anti-MYH14.. For blotting immunoprecipitated MYH14, NBP3-29780 was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Background: Myosin Heavy Chain

Myosin heavy chain is a muscle myosin, which is a hexameric protein that consists of 2 heavy chain subunits (MHC), 2 alkali light chain subunits (MLC) and 2 regulatory light chain subunits (MLC-2). Cardiac myosin heavy chain exists as two isoforms in humans, alpha-cardiac MHC and beta-cardiac MHC. These two isoforms are expressed in different amounts in the human heart. During normal physiology, beta-cardiac MHC is the predominant form, with the alpha-isoform contributing around only 7% of the total MHC. Mutations of the myosin heavy chain genes are associated with several different dilated and hypertrophic cardiomyopathies.
