Western Blot: Nicotinic Acetylcholine R alpha 1/CHRNA1 Antibody (mAb 192) [NBP2-81070] - Mouse skeletal muscle lysate (35ug protein in RIPA buffer) was resolved on a 10% SDS PAGE gel and blots probed with the chimeric mouse IgG1 version of mAb 192 at 0.03 ug/ml before detection using an anti-mouse IgG1 secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The predicted running size for AChR is 58.9 kDa. Chimeric mouse igG1 version of mAb 192 successfully detected AChR in mouse skeletal muscle tissue lysate. Immunocytochemistry/Immunofluorescence: Nicotinic Acetylcholine R alpha 1/CHRNA1 Antibody (mAb 192) [NBP2-81070] - Immunofluorescence analysis of paraformaldehyde fixed HeLa cells on Shi-fix coverslips, permeabilized with 0.15% Triton and stained with the chimeric mouse IgG1 version of mAb 192 at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2 ug/ml), showing cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81070, DAPI, merged channels and an isotype control. The isotype control was stained with an unknown specificity antibody followed by Alexa Fluor 488 secondary antibody.Applications
