Nup153 Antibody (8D4)

Immunohistochemistry: Nup153 Antibody (8D4) [NBP3-26450] -

Immunohistochemistry: Nup153 Antibody (8D4) [NBP3-26450] - Image of Nup153 Antibody (8D4) diluted at 1:100 and staining in paraffin-embedded human breast cancer performed. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4C overnight. The primary is detected by a Goat anti-rabbit IgG polymer labeled by HRP and visualized using 0.05% DAB.

Immunocytochemistry/Immunofluorescence: Nup153 Antibody (8D4) [NBP3-26450] -

Immunocytochemistry/Immunofluorescence: Nup153 Antibody (8D4) [NBP3-26450] - Staining of Hela Cells with Nup153 Antibody (8D4) at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeated by 0.2% Triton X-100, and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. Nuclear DNA was labeled in blue with DAPI. The secondary antibody was FITC-conjugated Goat Anti-Rabbit IgG (H+L).

Background: Nup153

Nuclear pore complexes are extremely elaborate structures that mediate the regulated movement of macromolecules between the nucleus and cytoplasm. These complexes are composed of at least 100 different polypeptide subunits, many of which belong to the nucleoporin family. Nucleoporins are pore complex specific glycoproteins characterized by cytoplasmically oriented O linked N acetylglucosamine residues and numerous repeats of the pentapeptide sequence XFXFG. Nup153 has three distinct domains: a N terminal region within which a pore targeting domain has been identified, a central region containing multiple zinc finger motifs, and a C terminal region containing multiple XFXFG repeats. Nup153 is a possible DNA binding subunit of the nuclear pore complex (NPC). The repeat containing domain may be involved in anchoring components of the pore complex to the pore membrane.
