Western Blot: Olig2 Antibody [NBP2-27301] - Mouse brain lysate was separated on a 12% gel using SDS-PAGE, protein transferred to PVDF membrane and blocked in 5% milk-TBST. The membrane was probed with 2 ug/mL Olig2 antibody in 1% milk and detected with an anti-rabbit HRP conjugated secondary using chemiluminescence.

Immunocytochemistry/Immunofluorescence: Olig2 Antibody [NBP2-27301] - Neuro2a cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with Olig2 NBP2-27301 at a 1:200 dilution overnight at 4 degrees Celsius and detected using Dylight 488 (Green). Alpha tubulin was used as a co-stain at a 1:1000 dilution and detected with Dylight 550 (Red). Nuclei were detected with DAPI (Blue). Cells were imaged using a 40X objective.

Background: Olig2

OLIG2 [oligodendrocyte (OL) transcription factor 2], an important regulator of OL development, is a bHLH (basic helix-loop-helix) expressed in both the developing and mature central nervous system (CNS). OLIG2 has critical functions at several stages of neural development including neuroepithelial patterning, neural cell progenitor fate specification and motor neuron differentiation (reviewed in Ono, 2009). OLIG2 positive lineage stem cells differentiate into neurons or glial cells, and it is tbought that OLIG2 may be required for OL differentiation and adult re-myelination. OLs, a type of glial cell, make and maintain myelin, a multi-layered glial sheath, in the CNS. OLIG2 is referred to an OL neural cell lineage specific differentiation marker because it is expressed in the neural progenitor cells that give rise to OLs during brain development as well as in mature OLs. OLIG2 is also important in the diseased (Zaidi et al, 2009) and injured brain (Ono, 2009). For example, it is thought that brain tumors may arise from brain tumor stem-like cells (Zaidi et al, 2009), and OLIG2 is a putative biomarker of diffuse gliomas including astrocytomas, oligodendrogliomas, and oliogoastrocytomas (Ligon et al, 2004). Diffuse gliomas are the most common primary brain tumors and tend to have high OLIG2 expression levels. Although the exact mechanism and identity of the cell(s) of origin of a glioma stem-like cell remains to be elucidated, OLIG2 may be required for tumor initiation. One theory is that OLIG2 triggers the proliferation of glioma cancer-initiating stem-like cells by suppressing p21, thereby lifting the growth inhibitory effects of p21 (reviewed in Li et al, 2009). Antibodies to OLIG2 are widely used as markers of OLs and their progenitors, as well as for identifying diffuse gliomas (Durand et al, 2010, Ligon et al, 2004, Marie et al, 2001, Yokoo et al, 2004).
